Metachromatic Effects on Interaction between Toluidine blue and L-a-Lecithin

Abstract
The absorption spectra of Toluidine blue(TB) in aqueous solution have been studied in tenns of monomer-dimer equilibria in the 10-6 to 10-4M concentration range. ¥ðle metachromatic behaviors of toluidine blue in the presence of various Lipid(L- ¥á -Lecithin) have been investigated by means of absorption spectroscopy. Toluidine blue is a metachromatic dye and is frequently employed in that capacity. It is a blue nucler counterstain and demonstrates nissl substance. It is used to stain mast cell granules, both metachromatically and orthochromatically.Bradley and Wolf¡¯s model of dye binding on polyanion. In this model dye molecules bound to anionic sites of the polyanion may be sufficiently closely located on the polyanion to interact with one another to from aggregates analogous to those found in free due solution. P/D is the ratio of concentration of polyanion available sites to bound dye concentration.The characteristic changes of metachromatic band with the change of L/D(lipidjdye) value are discussedin tenns of stacking theory.
